摘要
目的 检测 30例食管鳞状上皮细胞癌中IL -8和其它血管生成相关的化学因子的mRNA水平 ,对食管鳞癌新生血管进行定量分析 ,并分析IL -8与CD10 5血管平均距离的相关关系。方法 采用多探针RNA酶保护性分析检测IL -8和其它血管生成相关的化学因子的mRNA水平 ,免疫组织化学方法 (CD10 5)和Image -pro -plus软件进行新生血管的定量分析。结果 IL -8、IP -10和MIP -1α、βmRNA水平在食管癌组织中显著升高 ,Rantes、MCP -1无明显变化 ;在食管鳞癌CD10 5低血管密度病人 ,IL -8mRNA与CD10 5血管平均距离具有较好的相关性 (r=0 .6 2 ,P <0 0 5 ) ,在CD10 5高血管密度病人中无相关性 (r =0 .12 ,P >0 0 5 )。结论 IL -8在食管鳞癌血管生成过程中起重要作用。IL -8的促血管生成作用可能发生在食管鳞癌血管生成的早期阶段。MIP -1α、β和IP -10可能是食管癌血管生成过程中抑制肿瘤血管生成的重要调节因子。
Aim To detect the mRNA expression levels of interleukin-8 and other angiogenesis-related chemokines in 30 primary esophageal squamous cell carcinoma (ESCC) and adjacent normal tissues to quantify the activated vessel in EC tissue and analyze the correlation between IL-8 mRNA and CD 105 stained vessel distance.Methods Multiple-probe RNase protection assay was used to determine the mRNA expression.Activated vessel distance was quantified by immunohistochemical staining and image-pro-plus software.Results The mRNA expression of IL-8 ,IP-10 and MIP-1 α?β were significantly increased in ESCC compared with that of adjacent normal tissue,and no obvious changes were observed with Rantes and MCP-1.There was significant correlation between IL-8 mRNA and CD 105 stained vessel distance in patients with low vasculature (r=0.62,P<0.05),but not in those with high vasculature (r=0.12,P>0.05).Conclusions IL-8 may play an important role in primary esophageal carcinoma,and its function to promote tumor angiogenesis may be exerted in early stage.MIP-1 α?β and IP-10 may be important anti-angiogenesis factors.
