摘要
目的 从设计针对人TXSmRNA的核酶中 ,筛选能显著抑制TXSmRNA表达的核酶。方法 将核酶DNA亚克隆到真核表达载体pEGFP ,经脂质体转染入ECV30 4细胞后 ,用RT PCR方法测定细胞内TXSmRNA的相对含量。结果 核酶R4 16、R4 4 1能显著抑制ECV30 4细胞TXSmRNA的表达。结论 核酶可望成为人TXS有效的抑制剂 。
Objective To select effective hammerhead ribozymes to inhibit TXS mRNA in ECV304 cell line. Method The plasmids of pEGFP R were delivered into ECV304 cells by liposomes. The expression of TXS mRNA was investigated by RT PCR. Result R416?R441 could decrease TXS mRNA in ECV304 cells. Conclusion R416?R441 might be good inhibitors of TXS in gene therapy.
