DDPH对HECCM促PAS MC增殖后的逆转及时间相关性的研究

摘要 目的 研究DDPH对肺动脉平滑肌细胞 (PASMC)增殖后的逆转效应与DDPH的作用时间依赖性。方法采用细胞培养、四甲基偶氮唑盐比色法 (MTT)、免疫细胞化学染色测定α SM actin等方法观察低氧内皮细胞条件培养基(HECCM )促猪肺动脉平滑肌细胞的增殖作用 ,以及DDPH对肺动脉平滑肌细胞 (PASMC)增殖后的逆转效应与DDPH的作用时间依赖性。结果  (1)在HECCM促使PASMC增殖后 ,PASMC的表型发生转化 ,由收缩型转化为合成型 ,而DDPH能使HECCM促PASMC增殖后的表型 ,由合成型逆转为具有执行正常收缩功能的收缩型 ;(2 )在HECCM促PASMC增殖 2 4h ,36h ,4 8h后 ,各加入DDPH2 4h ,36h ,4 8h后 ,MTT法测定OD值下降 ;免疫组织化学测定α SM actin的吸光度上升 ,并具有时间依赖性。 Objective To study the reverse effect of DDPH on post proliferation PASMCinduced by HECCM and the time dependent manner of the effect. Method By using cell culture, MTT colorimetry andimmunocytochemistry, we observed the effects of HECCM on PASMC proliferation, the reverse effects of DDPH on the post proliferation PASMC and its time dependent manner Result (1) HECCM could promote PASMC proliferation and transform these cells from a contractile to a synthetic phenotype, while DDPH could reverse the proliferation, transformining them from a synthetic to a contractile phenotype; (2)after 24h, 36h and 48h ofPASMC proliferation which was stimulated by HECCM, DDPH was put in and the 3 PASMC groups were then cultivated for another 24h, 36h, 48h respectively; the results showed that the OD value decreased and the absorbance value increased Conclusion Our study suggests that DDPH could reverse PASMC proliferation stimulated by HECCM in a time dependent manner
