摘要
目的 了解血管紧张素 Ⅱ (Ang Ⅱ ) ,心钠素 (ANP)对人内皮细胞碱性成纤维细胞生长因子 (bFGF)表达的影响。方法 采用培养的人脐静脉内皮细胞 ,将Ang Ⅱ和ANP分别加入培养液内 ,通过免疫组织化学方法 ,原位杂交技术 ,结合图像分析法 ,测定各组内皮细胞bFGF蛋白及mRNA的表达变化。结果 Ang Ⅱ (10 8mol/L)对内皮细胞bFGF表达有明显促进作用 ,蛋白合成比对照组增加 5 9 3% (P <0 0 1) ;ANP (10 -7mol/L)对培养的内皮细胞bFGF表达有明显抑制作用 ,蛋白合成可减少 71 3% ;二者同时加入 ,蛋白合成比正常对照组减少 32 8% (P <0 0 1) ,各组间比较 ,差异具有统计学显著性意义 (P <0 0 1)。内皮细胞bFGFmRNA的变化与蛋白合成的改变基本相似。结论 ANP能拮抗AngⅡ对培养的人内皮细胞bFGF表达的促进作用 ,其作用可能是通过减少mRNA。
Objective To investigate the effects of angiotensin Ⅱ(Ang Ⅱ) and atrial natriuretic peptide (ANP) on the expression of basic fibroblast growth factor(bFGF) inhuman endothelial cells. Method Human umbilical vein endothelial cells were cultured in vitro and exposed to (1) Ang-II(10 -8 mol/L), (2)ANP(10 -7 mol/L), (3)Ang-II(10 -8 mol/L) plus ANP(10 -7 mol/L) , and(4)PBS, respectively The levels of bFGF expression in endothelial cells were determined by immunohistochemistry, in situ hybridization and image analysis technique Result The bFGF protein was distributed in the endothelial cell cytoplasm After the exposure to Ang Ⅱ, the bFGF expression increased by 59 3% as compared with that of the controls ( P <0 01); but the exposure to ANP decreased the bFGF expression by 71 3%( P <0 01) After the exposure to both, the bFGF expression decreased by 32 8%( P <0 01) There were significant statistical differences between every two groups( P <0 01) BFGF mRNA was distributed in the cytoplasm and nucleus, and the changes of bFGF mRNA contents were similar to those of bFGF protein Conclusion These results show that ANP, as an antagonist to Ang Ⅱ, can inhibit bFGF expression of endothelial cells through reducing bFGF mRNA
