活血潜阳胶囊对血管平滑肌细胞增殖的影响

Influence of "Blood-Activating and Yang-Suppressing Capsules" on Proliferation of Vascular Smooth Muscle Cells

为研究活血潜阳胶囊对血管紧张素Ⅱ (ATⅡ )诱导的血管平滑肌细胞 (VSMC)增殖的影响。以贴块法行SD大鼠胸主动脉VSMC分离培养 ,以血清药理学方法进行实验 ,10 - 6 MATⅡ为刺激因子 ,Captopril为对照 ,活血潜阳胶囊药物血清分为 10 %、7 5 %、5 %、2 5 % 4个浓度。绘制细胞生长曲线 ,并测定DNA的合成。结果 :经活血潜阳胶囊处理后 ,细胞增殖速度明显下降 ,生长曲线下移 ,3H -TdR掺入量减少 ,DNA合成明显受到抑制 ,且在一定范围内有剂量依赖性。提示 :活血潜阳胶囊能抑制ATⅡ引起的VSMC增生 ,揭示了活血潜阳胶囊治疗高血压的机理之一。

In order to study the influence of 'Blood Activating and Yang Suppressing Capsules' on vascular smooth muscle cells(VSMC)induced by angiotensin Ⅱ(ATⅡ),VSMC from the thoracic aorta of SD rats was separated and cultured with the strapping method for experiment by serological pharmacology,with 10 -6 M ATⅡ as the irritating factor,Captopril as the control and with medicated serum of 'Blood Activating and Yang Suppressing Capsules'in four concentrations of 10%,7 5%,5% and 2 5%,to draw the growth curve of the cells and determine DNA synthesis.The results showed that after management of 'Blood Activating and Yang Suppressing Capsules',the velocity of cell proliferation decreased obviously,with downward migration of the growth curve and reduction in 3H TdR mixture,and DNA synthesis was obviously inhibited,in dose dependence in certain extent.The findings indicate that 'Blood Activating and Yang Suppressing Capsules' can inhibit VSMC proliferation induced by ATⅡ,which shows one of the mechanism of hypertensive vessel of 'Blood Activating and Yang Suppressing Capsules'.
