摘要
目的 检测吸烟者外周血淋巴细胞 DNA的损伤状况 ,探讨单细胞凝胶电泳在分子流行病学研究中的应用价值。方法 单细胞凝胶电泳。结果 吸烟者的外周血淋巴细胞 DNA损伤显著重于非吸烟者 ,拖尾长度分别为 (2 7.5± 2 .8) μm和 (16 .2± 1.5 ) μm;尾矩分别为 11.3± 1.7及 3.0± 0 .9,并且呈现剂量反应关系 ;随着吸烟剂量的增加 ,外周血淋巴细胞的 DNA损伤加重。结论 吸烟可以造成外周血淋巴细胞 DNA损伤。单细胞凝胶电泳能够较灵敏地反映人群因有毒化学物质造成的 DNA损伤 ,在分子流行病学研究中具有重要的应用价值。
Objective In order to explore the biomarker of smoker, we have detected the DNA damage of peripheral blood lymphocytes (PBL).Methods Single Cell Gel Electrophoresis.Results The PBL DNA damage of smoker was more serious than that of non smoker, the tail lengths were (27.5±2.8)μm and (16.2±1.5)μm respectively, the TMs were 11.3±1.7 and 3.0±0.9 respectively, the difference was significant; and a dose effect relationship was observed.Conclusions Smoking can cause DNA damage of PBL, and SCGE can reflect the DNA damage caused by chemical poison. SCGE could be used in molecular epidemiological study.
