摘要
本文对鱼类人工雌核发育中两个技术处理进行了优化分析;对精子的照射时间、照射条件和卵子染色体加倍的温度休克等技术进行了最佳选择。目的在于推广应用于生产。在30w×2支紫外线灯、照射距离10cm、精液厚度0.2mm条件下,最佳照射时间为900秒左右;热休克和冷休克诱导鱼卵染色体加倍均可获得一定数量的雌核二倍体鱼;在生产上进行大量鱼卵处理时,鲤鱼采用4℃低温休克1,000秒、草鱼4℃低温休克900秒是可行的。
This article conducts an optimum analysis of two technical treatments in the artificial female nucleus development of fish and makes the best selection of the techniques of irradiating time and irradiating conditions of sperms and the thermal shock of ovum chromosome redoublement so as to popularize and use the techniques in production. Under 2 ultraviolet lamps of 30 w, 10cm irradiating distancs and 0.2mm thickness of milt, the optimum irradiating time is about 900 seconds, a certain number of diploid fish of female nucleus may be gained by the ovum chromosome redoublement induced by thermal shock and cold shock. When treating spawns in quantity in production, 4℃ low temperature shock to carp for 1,000 seconds and to grass carps for 900 seconds is feasible.
