摘要
以铜离子为模板定向合成了6-O-壳聚糖硫酸酯,然后分别用乙醛酸和丙酮酸在其N-位形成席夫碱,再经NaCNBH3还原制备了N-羧甲基-6-O-壳聚糖硫酸酯和N-(2-甲基-羧甲基)-6-O-壳聚糖硫酸酯。对制得的各化合物分别采用氧瓶燃烧法测定了硫含量(S%),采用HPGPC法测定了重均相对分子质量和相对分子质量分布宽度,并对各化合物的红外光谱(IR)和13C-NMR核磁共振图谱进行了分析。
6-Osulfochitosan (6S) was prepared regioselectively by using HCONH2/HSO3Cl method, following protection of the C2-NH2 group and C3-OH group of chitosan with copper ions(Ⅱ) at first, and then N-carboxymethyl-6-O-sulfochitosan and C3-OH group of chitosan with copper ions(Ⅱ) at first, and then N-carboxymethyl-6-O-sulfochitosan and N-(2-methyl-carboxymethyl)-6-O-sulfochitosan were obtained respectively by using glyoxylic acid mono-hydrate and pyruvic acid with 6-O-sulfochitosan to give a schiff base, then reduced by sodium cyanoborohydride at room temperature. Their contents of sulfur were determined by oxygen flask method, and their weight average molecular weight (Mw) and the distribution width of molecular weight (Mw/Mn) were determined by high performance gel permeation chroma-tography (HPGPC) respectively, and their 1R spectrum and 13 C-NMR spectrum were measured in this paper also.
