当归对大鼠缺血性脑损伤神经细胞黏附分子表达的影响 被引量:3

摘要 目的 :研究大鼠缺血性脑损伤后 ,当归对神经再塑过程中神经细胞黏附分子 (NCAM )表达的影响。方法 :雄性Wistar大鼠 5 0只 ,体重 170— 2 0 0 g ,随机分为缺血损伤组 (n =2 5 )和当归治疗组 (n =2 5 )。采用线栓模型造成右侧大脑中动脉阻断(MCAO) ,治疗组腹腔注射当归注射液 ,剂量为 5 g/kg。测定其中 10只动物的局部脑血流量 (regionalcerebralbloodflow ,rCBF)。分别在再灌注后 1d、2d、3d和 7d取材 ,观察大脑表面的血管形态 ,用免疫组织化学的方法 ,检测脑片NCAM的表达。结果 :与缺血损伤组 4个时间点相比 ,当归治疗组的rCBF显著增加 ,颞叶皮质NCAM的表达量显著增加。结论 :大鼠短暂缺血性脑损伤后 。 Objective: To investigate the effects of Angelica sinensis on the expression of neural cell adhesion molecule (NCAM) on the neural plasticity after the ischemic brain injury in rat. Method: Fifty male Wistar rats with an average body weight of 185±15 g (mean±SD) were used, and were randomly divided into two groups: group A rats (n=25) underwent middle cerebral artery occlusion (MCAO) for 2 hours by suture, group B rats (n=25) underwent MCAO for 2 hours meanwhile received treatment with Angelica sinensis (5 g/kg, i.p.). The regional cerebral blood flow (rCBF) in 4 of group A rats and 6 of group B rats were respectively measured. The brains were removed at 1 day, 2 days, 3 days and 7 days after reperfusion. The morphology of cerebral blood vessels was observed. The expression of NCAM in brain sections was measured by immunohistochemistry. Result: Comparing with group A, the rCBF in the group B significantly increased, and the expression of NCAM in the temporal lobe in the group B at four time points significantly increased respectively. Conclusion: The increasing amount of NCAM expression was enhanced by Angelica sinensis following transient interruption of cerebral blood flow in rats.
