微生物发酵生产十三碳二元酸的研究

摘要 以一株热带假丝酵母(Candida tropicalis)TSW_1为出发菌株,选用不同种类、不同能量、不同剂量的离子进行离子束注入处理,得到从十三碳烷烃生产十三碳二元酸的突变菌株TSW-N-35。研究了N^+和CH_3^+离子注入剂量与Candidda tropicalis存活率之间的关系。菌株的存活率均随着注入剂量的变化表现为相似的“马鞍型”,以十三碳烷烃为底物,在摇瓶条件下,十三碳二元酸(DCA13)产量达71.5g/L。研究发现,蔗糖浓度显著地影响菌体生长,其最适浓度为0.3%;尿素和丙烯酸的浓度显著地影响DCA13的产量,尿素的最适浓度为0.14%;添加0.10%丙烯酸,DCA13的产量能提高20%左右; A mutant. Candidda tropicalis TSW-N-35. which can produce 1,13 - tridecandioic acid from n-tridecane, was obtained after ion implanted with different variety, energy and dose, take TSW-1 as primary strain. The relations between the survival and dose of Candidda tropicalis by N+ and CH3+ ion implantation were investigated. The curves of survival all took a ' saddle shape with increasing ion implanted dose . On properly condition, take n-tridecane as base material, the amount of 1.13 -tridecandioic acid was up to 71.50g/L. It was found that the concentration of sucrose remarkably affected the strains growth, its optimal concentration was 0.3% . The yield of DCA13 was remarkably affected by the concentration of urea and acrylic acid. The optimal concentration of urea was 0.14% . The addition of 0.1% acrylic acid could increase the yield of DCA13 about 20% . The fermentation time could shorten by increase of the cultural temperature.
