摘要
对柑橘原生质体的超低温保存进行研究 ,发现玻璃化溶液加入小牛血清蛋白对原生质体的超低温保存有利 ,不同品种的胚性愈伤组织原生质体超低温保存后的成活率不同 ;不同倍性的伏令夏橙原生质体超低温保存后的成活率也不同 ,其中 2倍体成活率最高 ,其次为 4倍体 ,6倍体成活率最低 .超低温保存后的原生质体在BH3培养基中能够恢复分裂 。
A study was conducted to cryopreservation of citrus protoplasts. Results showed that bovine serum albumin added in PVS was benefit to cryopreservation of protoplasts. Protoplsats derived from different cultivars embryogenic calli had different survival rate after cryopreservation. The survival rates of different ploidy citrus sinensis (L.) Osbeck cv. valencia protoplasts were varied, among them, the survival rate of diploid cells was the highest and that of the ceus hexploid was the lowest. Protoplasts after cryopreservation could divide in BH 3 medium and could form cell colonies.
