摘要
报道了火炬姜叶片离体培养的研究结果 ,试验表明 :诱导叶片产生愈伤组织效果较好的激素配比是 6 -BA 0 .5mg·L- 1+ 2 ,4 -D 0 .5mg·L- 1;最适于愈伤组织分化的基本培养基是MS ;6 -BA、ZT和KT均能诱导愈伤组织分化不定芽 ,其中以 6 -BA的效果较好和经济 ;在所试的 6 -BA质量浓度中 ,以 3mg·L- 1的效果较佳 ;添加 80 0mg·L- 1的CH有利于火炬姜愈伤组织的分化 ;生根培养阶段 ,以附加w =0 .15 %的活性炭的 1/ 2MS培养基效果较好 ,生根率达 10 0 % ,且植株生长健壮 ;试管苗移栽在表土上 ,成活率可达 10 0 %
The research results of in vitro culture of Phaeomeria magnifica leaf is presented in this paper. The experiments demonstrate that: the optimal hormone ratio for inducing leaf callus is 6-BA 0.5 mg·L -1 +2,4-D mg·L -1 ; the optimal medium for callus differentiation is MS medium; 6-BA, ZT and KT can all induce callus to differentiate adventitious shoots, among them 6-BA is better and more economical; among all tested 6-BA mass concentraton, 6-BA of 3 mg·L -1 has a good effect; adding 800 mg·L -1 CH is good for callus differentiation; in rooting culture phase, 1/2 MS medium with w= 0.15% activated carbon has a good effect, the rooting rate can reach 100%, and the plants grow robustly; when planted in superficial soil, the tissue culture shoots have a survival rate of 100%.
