人滑膜间充质干细胞向软骨诱导分化的基础研究

摘要 目的 探讨人滑膜间充质干细胞(SMSCs)向软骨诱导分化的条件及分子机制,为半月板组织工程修复提供基础。方法 选取膝关节创伤患者5例,关节镜下取滑膜组织4~8片,每片约1 mm×3 mm×1 mm,加入含10%胎牛血清的低糖培养基进行原代及传代培养,以单克隆培养法获得纯化的滑膜间充质干细胞系,后采用流式细胞仪进行干细胞表型鉴定,并向完全培养基加转化生长因子-β1(TGF-β1)、地塞米松(DEX)、维生素C(VC)进行软骨诱导分化,采用免疫组化法检测诱导后Ⅱ型胶原(COLⅡ)的表达情况。结果 含10%胎牛血清的低糖DMEM培养基最有利于细胞生长,采用单克隆培养法可获得纯化较高的细胞;流式鉴定细胞表型:细胞表达CD44、CD90,不表达CD34、CD45;免疫组化法检测未经诱导的细胞形态基本保持梭形,而诱导14 d后逐渐由小梭形变为多角形,可见大量棕色颗粒,COLⅡ阳性表达。结论 人SMSCs优化生长条件:含10%胎牛血清的低糖DMEM培养基最有利于细胞生长,采用单克隆培养法可获得纯化较高细胞,且分离纯化的细胞免疫原性较低,适用于同种异基因体之间移植,可作为半月板组织工程修复的种子细胞。
