摘要
目的 从女贞叶有效部位皂甙中提取有效成分熊果酸 ,鉴定其化学结构 ;检测其对牙周卟啉菌、中间型普氏菌和福赛氏类杆菌的作用。方法 按照中药硅胶柱层析的方法 ,从皂甙中分离出晶体Ⅰ ,对晶体Ⅰ进行红外光谱、1 3C和1 H核磁共振谱分析 ;利用微量液体稀释法测定晶体Ⅰ对 3种牙周病原菌的最小抑菌浓度和最小杀菌浓度 ;同时测定晶体Ⅰ对中间型普氏菌和福赛氏类杆菌的杀菌曲线。结果 晶体Ⅰ经过波谱分析鉴定其结果为熊果酸 ,熊果酸对 3种牙周病原菌的最小抑菌浓度为 0 740 μg/L ,最小杀菌浓度为 0 2 95μg/L ,杀菌曲线显示 0 80 0 μg /L的熊果酸可在 3h内杀灭中间型普氏菌 ,6h内杀灭福赛氏类杆菌。
Objective To extract the effective ingredient(crystal Ⅰ) from effective section (saponin) of Ligustrum Lucidum Ait, identify the chemical structure of crystal Ⅰ, study the effect of crystal Ⅰ on P gingivalis, B forsythus and P intermedia Methods Isolated crystal Ⅰfrom saponin using the silica gel column chromatograph Identified crystal Ⅰ with IR spectra, 1H NMR and 13 C NMR Measured the minimal inhibitory concentration (MIC) and the minimal bactericidal concentration (MBC) through micro liquid dilution Studied the killing curve of ursolic acid on B forsythus and P intermedia Results The crystal Ⅰ was identified as ursolic acid; its MIC and MBC to P gingivalis, B forsythus and P intermedia were 0 740 and 0 295 μg/L respectively The killing curve indicated that 0 800 μg /L ursolic acid could kill P intermedia and B forsythus in 3 and 6 hours respectively Conclusion Ursolic acid has obvious effect to inhibit periodontal pathogen.
