透明质酸酶对人乳腺癌细胞MDA231增殖和转移的影响 被引量:1

摘要 目的 观察透明质酸酶 (PH 2 0 )基因对人乳腺癌细胞在鸡胚胎中生长和转移的影响。方法 PH 2 0cDNA转染到人乳腺癌细胞株MDA2 31中 (MDA2 31 PH2 0 ) ,细胞种植到鸡胚绒毛尿囊膜 (CAM )上 ,观察肿瘤大小 ;对肿瘤组织血管内皮细胞染色 ,研究肿瘤新生血管生长变化 ;PCR扩增远端CAM中AluDNA序列 ,观察PH \| 2 0对肿瘤细胞转移的影响。结果 MDA2 31 PH2 0在CAM上增殖的程度明显高于对照组 ,血管内皮细胞数量也增多 ,同时远端CAM出现AluDNA序列 ,而对照组无。结论 PH 2 0明显促进癌组织内新生血管生长 ,并促进乳癌细胞增殖和远端转移。 Objective To study the effects of human sperm membrane\|bound hyaluronidase (PH\|20) on gene transfected MDA231 cells growth and metastasis.Methods PH\|20 cDNA was transfected into human breast cancer cell line MDA231.Transfectants (MDA231\|PH\|20) were then implanted onto chorioallantoic membrane (CAM). Meanwhile, empty vector pcDNA3 was transfected into MDA231 (MDA231\|pcDNA3) as control. The followings were performed for the tumor growth and metastasis study: (1) weighting and measuring the tumors; (2) endothelial cells staining for angiogenesis;(3) PCR amplifyingAlu DNA of remote CAM tissue. Results Comparing to MDA231\|pcDNA3, the MDA231\|PH\|20 exnografts were larger and the level of angiogenesis was increased. Alu DNA was appeared on low level CAM and no Alu was found in control group.Conclusion The results suggested that PH\|20 may increase the palingenesis of blood vessel and stimulate human breast cancer cell growth and metastasis.
