IL-1对多发性骨髓瘤细胞粘附分子表达的影响

摘要 为探讨人骨髓瘤细胞株KM3表面粘附分子的表达及IL-1对粘附分子表达的影响,应用直接免疫荧光标记流式细胞术分析人骨髓瘤细胞株KM3细胞表面粘附分子表达,以荧光阳性率和平均荧光强度两个指标反映粘附分子的表达量,研究IL-1对KM3细胞粘附分子表达调控。发现正常情况下,KM3细胞表面表达粘附分子CD44、CD54、CD49b、CD49e、CD49f,不表达CD56和CD49d;IL-1以剂量非依赖方式促进KM3细胞表面CD44、CD54、CD49b、CD49e、CD49f表达。 Adhesion molecule expression of human myeloma cell line KM3 was examined by direct immunofluorescent flow cytometry analysis. KM3 cell surface mainly expressed CD 44, CD 54, CD 49 b, CD 49 e and CD 49 f (positive 20%~97%), and lacked CD 56 and CD 49 d expression (positive <5%). When exogenic IL-1 was added in the KM3 culture, expressions of CD 44, CD 40 b, CD 49 e, CD 49 f and CD 54 were increased, while CD 49 d and CD 56 did not change significantly (P>0.05). No significantly difference of KM3 cell surface adhesion molecule expression was found in various levels of IL-1. These results showed that IL-1 can promote adhesion molecule expression with dose independent pattern.
