粘附细胞作为APC提高MS淋巴细胞对髓鞘抗原增殖反应的探索

摘要 目的 比较粘附细胞作为APC能否较一般方法提高T细胞对神经髓鞘类抗原的增殖反应性和显示出MS对神经髓鞘类抗原的特异自身免疫反应特征。方法 选HLA DR15亚型MS和正常人外周血单个核细胞 ,体外培养。分别用一般T淋巴细胞增殖试验和富集粘附细胞法检测对人神经髓鞘、蛋白脂蛋白 (PLP)和碱性髓鞘蛋白(MBP)淋巴细胞增殖反应性。结果 富集粘附细胞法使MS组和对照组T淋巴细胞对神经髓鞘、PLP和MBP的增殖较标准法提高 (P <0 .0 5 ) ;使MS组以MBP和PLP的增殖反应较对照组的差别具有显著性的意义 ;而对TT无明显提高作用。结论 富集粘附细胞法可提高T淋巴细胞对神经髓鞘、PLP和MBP的增殖反应性 (MS和NC) ,多数情况使MS组和NC组的差别具有显著性的意义 ,对非髓鞘类抗原 (如TT)无这种作用。 Objective To find out if enriched adherent cell proliferation assay showed a higher degree of respose to myelin than mixed cell population.Methods Human T lymphocyte,from mononuclear cells of HLA DR15 type,proliferation assay was performed in both regular method and enriched adherent cell to myelin,myelin basic protein(MBP), proteolipid protein(PLP) and tetanus(TT),and then compare their results between two methods and two groups.Results Enriched adherent cell proliferation assay showed a higher degree of increased T cell proliferative respose to myilin,MBP and PLP,but not to TT than mixed cell population in MS or normal control(NC) and make the difference of proliferative respose to MBP and NC stronger.Conclusion Adherent cell as antigen presenting cell(APC) for enlipidated antigens such as myelin,not to TT showed a higher degree of respose than mixed cell population. Our results also suggested that myelin,MBP and PLP might play some important role in initiating demyelinating diseases.
