噬菌体肽库中筛选NMDA受体模拟抗原

摘要 [目的 ]研究发现脑缺血患者体内存在N甲基 -D -门冬氨酸 (NMDA)受体自身抗体 ,了解其滴度与疾病程度的相关性。 [方法 ]用NMDAR1单克隆抗体淘筛噬菌体展示随机 12肽库 ,获得NMDAR1模拟表位抗原。 [结果 ]细胞竞争ELISA结果显示 ,该模拟抗原可以竞争细胞表面天然抗原与抗体的结合 ,具有抗原模拟性。 [结论 ]噬菌体肽库中筛选NMDAR1受体模拟抗原可以用于临床病人血清检测。本研究为下一步临床检验准备了必要条件。 To determine the minotope of NMDAR1. The monoclonal antibody against the NMDAR1 was used as target protein to screen the binding peptide from a 12-mer Ph.D. random peptide library. After three rounds of affinity screening,the peptide sequences of positive phage clones were determined and analyzed by DNA sequencing,ELISA and competitive inhibition. A positve clone was found. It could competitively inhibit the native antigen binding to mAbN1. [Conclusion] This peptide mimics native NMDAR1 and may be useful for NMDAR1 autoantibody diagnosis in the patients with chronic cerebral circulation disorder.
