氢化可的松和胰岛素对兔软骨细胞生长的影响 被引量:3

摘要 目的 :观察胰岛素和氢化可的松对软骨细胞生长的影响。方法 :取新西兰兔膝关节软骨细胞 ,分别加入胰岛素、氢化可的松或二者的复合物的培养液中培养 ,用MTT法测定其对软骨细胞增殖的影响。结果 :单独使用胰岛素0 .0 35 μg/ml即可显著促进软骨细胞增殖 (P <0 .0 1) ,当浓度增至 0 .35 μg/ml时 ,其促进作用达到最大值 (P <0 .0 1)。氢化可的松 10 μg/ml时对软骨增殖的抑制作用明显 (P <0 .0 5 )。随着氢化可的松浓度的增大 ,抑制作用更显著 ,当浓度增至 10 0 μg/ml时 ,软骨细胞几乎全部死亡 (P <0 .0 1)。当胰岛素 0 .35 μg/ml与氢化可的松 5 0 μg/ml联合使用时 ,氢化可的松可以拮抗胰岛素的作用。结论 :低浓度胰岛素对软骨细胞的生长有促进作用。氢化可的松对软骨细胞的生长有抑制作用。二者联合使用时 ,氢化可的松可以拮抗胰岛素的作用。 Objective: To observe the effects of insulin and hydrocortisone on the proliferation of chondrocytes.Method:Chondrocytes were isolated and subcultured from the knees joints of New Zealand white rabbits,while insulin,hydrocortisone or insulin and hydrocortisone were added to cell culture media,the cellular proliferation was assayed by MTT method.Results:insulin could enhance the proliferation of chondrocytes at the concentration of 0.035μg/ml (P<0.01),and the enhancement reach the maximum at the concentration of 0.35μg/ml(P<0.01).10μg/ml hydrocortisone inhibited the proliferation of chondrocytes significantly (P<0.05),as the concentration increased to 100μg/ml,no viable chondrocytes were noticed.When the chondrocytes were treated with 0.35μg/ml insulin and 50μg/ml hydrocortisone,the action of insulin was inhibited.Conclusions:insulin at low concentration enhanced the proliferation of the chondrocytes,but hydrocortison could inhibit the growth of the chondrocytes.However,combinations of insulin and hydrocortisone showed that the action of insulin was inhibited by hydrocortison.
