反相高效液相色谱法测定夏天无中原阿片碱的含量

Determination of Protopine in Rhizoma Corydalis Decumbentis by RP-HPLC

目的 :建立用反相高效液相色谱 (RP- HPLC)法测定夏天无中原阿片碱含量的方法。方法 :采用 Shim Pack CLC-ODS柱 ,乙腈 -甲醇 - 0 .2 %磷酸溶液 (1 50∶ 1 0 0∶ 750 ) ,用三乙胺调节 p H值至 3为流动相 ,检测波长 2 82 nm。结果 :原阿片碱在 0 .60 6～ 3.0 30μg范围内与色谱峰面积呈线性关系 ;回归方程 :Y=39963.4X +2 61 .2 ,r=0 .99996;平均加样回收率及 RSD分别为 99.56%和 0 .92 %。结论 :该法可用于测定夏天无中原阿片碱的含量。

Objective:To establish the method for determining the content of protopine in Rhizoma corydalis Decumbentis by RP-HPLC.Methods:Using Shim Pack CLC-ODS column, acetonitrile methanol 0.2% phosphoric acid solution(150∶100∶750), then regulate pH to 3.0 with triethylamine as the mobile phase and detection wavelength at 282 nm.Results:The calibration curve was linear at a range of 0.606～3.030 μg for the protopine and the linear equation was Y =39 963.4 X +261.2, r =0.999 96. The average recovery was 99.56% and the related standard deviation ( RSD ) was 0.92%.Conclusion:This method can be used to determine the content of protopine in Rhizoma Corydalis Decumbentis .
