脑胶质瘤多药耐药基因表达及其对化疗药物敏感性 被引量:7

EXPRESSION OF MULTIDRUG RESISTANCE GENE IN BRAIN GLIOMA CELLS AND ITS SENSIBILITY TO CHEMOTHERAPEUTIC DRUGS

摘要 ①目的 检测 1型多药耐药基因 (MDR1)产物P170在人脑胶质瘤组织中表达及其对化疗药物敏感性。②方法 采用免疫组化的方法 ,对 38例人脑胶质瘤手术切除标本的MDR1表达水平进行检测 ;采用四唑蓝快速比色法检测氟尿嘧啶 (FU )、顺铂 (DDP)、甲氨喋呤 (MTX)、丝裂霉素 (MMC)、司莫司汀 (MeCCNU)、洛莫司汀(CCNU)、双氯乙基亚硝脲 (BCNU)、威猛 2 6 (VM 2 6 )对脑胶质瘤细胞的抑制率。③结果  38例人脑胶质瘤细胞P170阳性表达率为 6 5 .7% ,与正常脑组织 (表达率为 0 )比较差异有显著性 (P =0 .0 0 0 2 ) ;BCNU ,MeCCNU ,CCNU对胶质瘤的抑制率较高 ,FU ,MTX ,MMC对胶质瘤的抑制率较低 ,差异有显著性 (F =4 7.6 ,q =5 .2 5~ 6 .78,P <0 .0 5 )。脑胶质瘤对 8种化疗药物的敏感性差异有极显著性 ;随着胶质瘤恶性程度的增高 ,P170的阳性表达率逐渐升高 (胶质瘤Ⅰ~Ⅳ级的表达率分别为 0 ,5 3.3% ,70 .6 % ,10 0 .0 % )。④结论 脑胶质瘤的耐药性与MDR1表达水平密切相关。 Objective\ To determine the expression of P170 in brain glioma cells and to explore the relationship between the expression and the sensibility to chemotherapeutic drugs. \ Methods\ The levels of multiple drug resistance (MRD 1) expression in 38 cases of brain glioma were studied immunohistochemically. The inhibitory rates of FU, DDP, MTX, MMC, MeCCNU, CCNU, BCNU, VM 26 to brain gliomas were detected by tetrazolium base decoloimetric assay (MTT assay). \ Results\ The positive expression rate of 38 cases of brain glioma was 65.7%, which was significantly different from that of the normal brain tissue ( P =0.000 2). The inhibitory rates of BCNU, MeCCNU and CCNU were significantly higher than those of FU, MTX and MMC ( F=47.6, q=5.25-6.78, P <0.05). The difference of the sensibility of the eight chemotherapeutic drugs to gliomas was remarkable. The positive expression rate of P170 increased along with the development of the tumor grade, the rates being 0, 53.3% , 70.6% and 100.0% for grades Ⅰ to Ⅳ respectively. \ Conclusion\ The drug resistance in brain glioma is closely correlated with the expression level of MDR 1.
