天然生物膜对UVA辐射无毛小鼠皮肤的保护作用 被引量:6

摘要 ①目的 探讨天然生物膜 (NBM )对长波紫外线 (UVA)辐射所致小鼠皮肤氧化损伤的保护作用。②方法 建立UVA(辐射强度为 15 1.6 8J/cm2 ,共 30d)辐射昆明种无毛小鼠皮肤氧化损伤模型 (UVA模型 )。将无毛小鼠随机分为双蒸水未辐射组和UVA辐射组 (双蒸水辐射对照组、10 0 g/LNBM组、10 0g/L维生素C组 )。电镜观察皮肤组织超微结构 ;免疫组织化学方法测定小鼠表皮细胞的B细胞淋巴瘤 白血病 2 (Bcl 2 )蛋白表达、一氧化氮合酶 (NOS)的活性 ;生物化学法测定皮肤匀浆上清液中抗氧化酶 (GSH Px ,SOD)活性和丙二醛 (MDA)的含量及总抗氧化能力 (T AOC)。③结果 UVA辐射组小鼠皮肤的表皮细胞内可见空泡形成 ,真皮的成纤维细胞内可见大液泡形成。NBM组表皮细胞结构正常 ,成纤维细胞的粗面内质网丰富 ,NBM可以增强小鼠表皮细胞内Bcl 2蛋白的表达 ,并抑制NOS的免疫活性 ;NBM可提高小鼠皮肤组织的T AOC和SOD活性 ,降低MDA含量 ,与维生素C的抗氧化作用一致。④结论 天然生物膜具有抗UVA氧化损伤的作用 ,其作用机制可能与其增强Bcl 2的蛋白表达 ,降低NOS的活性及提高抗氧化酶活性有关。 Objective\ To study the protective effects of the natural biological membrane (NBM) on hairless mice damaged by ultraviolet A (UVA). \ Methods\ The oxidation damaged models of hairless mice irradiated by ultraviolet A (irradiation intension: UVA 151.68 J/cm 2 for 30 days) were established. The mice were randomly divided into control and UVA radiated groups ( Water model, 100 g/L NBM, 100 g/L Vit C). The ultrastructure of skin was observed under electron microscope. The expressions of Bcl 2 protein and NOS were examined by immunohistochemical method. The contents of MDA, the activities of GSH Px, SOD and the T AOC in the supernates of the mouse skin were detected by biochemical method.\ Results\ Dilated vacuolations and few rough endoplasm reticulum in fibroblasts and epidermic cells of mice in UVA model group were observed under electron microscope . The ultrastructure of epidermis and fibroblasts in the mice of NBM groups remained normal. NBM could enhance the expression of Bcl 2 protein and inhibit the activities of NOS, decrease the amounts of MDA and increase the contents of SOD and T AOC in the skin of the mice irradiated by UVA. \ Conclusion\ The NBM has the protective effects on the hairless mice irradiated by ultraviolet A. The mechanism of this protective effects of NBM may be mediated by enhancing the expression of Bcl 2 protein, inhibiting the activity of NOS, and increasing the activities of anti oxidative enzymes.
