摘要
随着分子生物学技术的进步,单克隆抗体的生产技术逐渐完善,但在生产中要制备出高质量的单抗仍存在许多问题。本文就动物的免疫、杂交瘤细胞的建立、融合细胞的筛选、克隆化培养及细胞冷藏、单克隆抗体的大量生产及纯化、单克隆抗体制备过程中的有关污染问题进行论述,以期对生产有一定的指导。
With the advancement of molecular biological technology, production of mono-cloned antibody is being improved. However many difficulties still exist in producing mono-cloned antibody of high quality. This paper covers animals' immunity, hybrid tumor cells, selection of merged cells, culture of cloning, refrigeration of cells, mass production and purification of mono-cloned antibody, and pollution during the production.
