GCNT3在结直肠癌中的表达及临床意义

Expression and Clinical Significance of GCNT3 in colorectal cancer

目的探讨GCNT3在结直肠癌中表达的水平变化及其对结直肠癌患者预后的影响。方法国际癌症和肿瘤基因数据库(TCGA)中下载结直肠癌患者GCNT3基因(编码C2GnT-2)的mRNA表达数据及临床信息,整理分析其表达水平变化对结直肠癌临床相关性及预后的影响,并通过细胞增殖实验观察干扰C2GnT-2合成后结直肠癌细胞增殖能力的变化情况。结果 GCNT3在癌组织中较癌旁组织低表达(P<0.001);GCNT3低表达组病人无病生存率显著低于高表达组(P<0.05);干扰C2GnT-2合成后结直肠癌细胞增殖能力显著增强(P<0.01)。结论 GCNT3在结直肠癌中低表达,其低表达可能提示直肠癌患者无病生存率低。

Objective To investigate the expression level of GCNT3,a glycosyltransferase enzyme,in colorectal cancer(CRC)and its association with the prognosis of the patients.Methods The mRNA expression data of GCNT3 gene(encoding mucin-type core 2 synthase enzyme,C2 GnT-2)and clinical information of the patients were obtained from the Cancer Genome Atlas database(TCGA).The correlation of GCNT3 mRNA expression level with the clinical characteristics of the patients was analyzed.Kaplan-Meier analysis and Cox regression analysis were used for assessing the prognostic value of GCNT3.We also tested the effect of a short hairpin RNA(shRNA)targeting GCNT3 on the proliferation of the colorectal cancer cell line HT-29 using MTT assay.Results GCNT3 mRNA expression level was significantly decreased in CRC tissues as compared with the paired adjacent colorectal tissues(P<0.001).A low expression level of GCNT3 was significantly correlated with a reduced disease-free survival(DFS)rate in the patients with CRC(P<0.05).In HT-29 cells,interference of C2 GnT-2 with the specific shRNA significantly promoted the cell proliferation as compared with the control cells(P<0.01).Conclusion The expression of GCNT3 is significantly downregulated in CRC tissues,and a low expression of GCNT3 can be indicative of a low disease-free survival rate of patients with CRC.
