摘要
本文涉及组织培养领域的一种新技术。它的基本特征是试管苗或脱毒苗生长在特制的敞口容器中。培养基和外植体由一层粉末材料覆盖,它可以有效地阻止微生物污染而不阻止脱毒苗茎叶的生长和钻出粉末层,阻止水分外渗而不阻止氧气渗入。由于茎叶暴露于容器外部,接受自然光的直射和干燥通风等条件的锻炼,试管苗生长矮壮、色绿,不用练苗而移栽成活率很高,其素质接近常规繁殖的幼苗。暴露培养法可以克服传统组培技术由于封闭系统方式而造成的幼苗娇弱,移栽后成活率低以及成本过高等固有缺点。
Plant tissue culture as a mass propagation technique for many commercially important plants is badly limited by its higher cost of production than that of the common nursery met(?)od. The main reason for the higher cost is the tenderness of the seedlings derived in vitro that are too delicate to survive after being transplanted out from the test tube or other containers. A new method is reported in the present paper that can efficiently overcome the tenderness and greatly increase the survival rate. The seedlings produced by the new method show thicker epidermal cuticle, smaller stomata guard cells and more epidermal hairs than those produced by the conventional method of plant tissue culture.
