摘要
目的探讨卵巢切除是否导致大鼠骨髓上清骨吸收活性增加,以及白细胞介素-6(IL-6)在骨吸收中的作用。方法通过胎鼠颅骨 45Ca释放实验测试骨髓上清骨吸收活性及IL-6抗体的中和作用,通过 ELISA检测骨髓上清 IL-6的含量。结果卵巢切除组 (OVX)骨吸收活性较对照组 (Sham)显著性升高 (P< 0.01),雌激素治疗组 (OVX+ E2)骨吸收活性较 OVX组显著性下降 (P< 0.01);IL-6抗体使 OVX组骨髓上清的骨吸收活性下降 63%。 OVX组骨髓上清 IL-6含量是 Sham组的2倍 (P< 0.01),OVX+ E2组较 OVX组下降了 30% (P< 0.01)。结论卵巢切除导致内源性骨吸收因子 IL-6产生增加,并且 IL-6在骨吸收增加中起关键作用,雌激素抗骨吸收作用部分是通过降低 IL-6产生而介导的。
Objective To export whether the ovariectomy induced increase in bone resorbing activity present in supernatants fractin of bone marrow and the effects of endogeneous IL-6 on bone resorption.Method The bone resorbing activity present in marrow supernatants and the neutralizing effects of antibody against IL-6 were examined by the release of 45Ca from perlabele fetal rats parietal bone,the concentration of IL-6 were measured by Enzyme Linked Immunosorbent Assay(ELISA).Result Adding bone marrow suqernatants at10% 60% to organ cultures of rats parietal bone dose dependently stimulated bone resorption.In same concentration condition the endogenous bone resorbing activity present in bone marrow supernatants from OVX rats were much higher compared with from Sham rats 8 weeks after surgery,and in OVX+ E2 group it were much lower compared with in OVX group.Anti IL-6 body neutralized significantly the bone resorbing activity present in bone marrow supernatants from OVX rats.The IL-6 concentration of bone marrow supernatants was much higher in OVX rats compared with in Sham operated rats,while its enhancement were suppressed completely by estrogen replacement therapy.Conclusion Estrogen deficiency after ovariectomy increased in bone resorbing activity present in bone marrow supernatants and it were mediated partialy by endogeneous IL-6,Suppression for IL-6 production were involvement in the mechanism of estrogen replacement therapy inhibiting bone loss in estrogen deficiency.
