摘要
目的:研究P210阳性慢性髓系白血病(chronic myeloid leukemia,CML)患者骨髓中单个核细胞Notch1和ASB2基因mRNA的表达,探讨P210阳性CM L患者中Notch信号通路与蛋白泛素化作用的相关性,为研究白血病发生发展的分子机制和临床治疗提供有价值的参考。方法:收集初诊的32例P210阳性CML患者以及34例非白血病和健康人(对照组)骨髓,采用实时荧光定量PCR检测骨髓中Notch1和Asb2 mRNA表达情况。结果:CML患者骨髓中Notch1 mRNA表达量相对于对照组升高了337.8倍,差异有统计学意义(t=36.3,P<0.01),CML患者骨髓中ASB2 mRNA表达量高于对照组(t=19.4,P<0.01)。Notch1与ASB2 mRNA表达量具有相关性(r=0.504,P<0.01)。结论:P210阳性CM L患者骨髓中可能存在Notch1和ASB2的异常表达,这种异常表达可能参与CM L的发生发展。
Objective: To explore the mRNA expression level of Notch1 and ASB2 in bone marrow mononuclear cells of patients w ith P210+chronic myeloid leukemia and the correlation betw een Notch signaling pathw ay and ubiquitination in chronic myeloid leukemia,so as to provide the valuable information for investigating the pathogenesis of chronic myeloid leukemia and clinical treatment. Methods: Bone marrow w as collected from 32 patients w ith new ly diagnosed chronic myeloid leukemia and 34 non-hematopathic and healthy individuals( control group),respectively. The mRNA expression levels of Notch1 and ASB2 w ere detected by real-time quantitative PCR. Results: The expression of Notch1 mRNA in CM L patients w ere significantly different from that of healthy individuals group( t = 36. 3,P < 0. 01),w hich w as 337. 8 times of the healthy individuals. M oreover,the expression level of ASB2 mRNA in CM L group w as significantly higher than that in healthy individuals( t = 19. 4,P < 0. 01). The mRNA expression of Notch1 and ASB2 gene w as positive correlation( r = 0.504,P < 0. 01). Conclusion: The aberrant expression of Notch1 and Asb2 exists in patients w ith P210 positive CM L,w hich may be involved in incidence and development of CM L.
