摘要
目的:探讨骨髓增生异常综合征(MDS)患者骨髓基质细胞IL-32 mRNA的表达及其与骨髓单个核细胞凋亡的相关性。方法:收集山东大学附属济南市中心医院26例MDS患者和10例缺铁性贫血(IDA,对照组)患者骨髓液,分离骨髓单个核细胞,分离培养骨髓基质细胞,采用RT-PCR检测骨髓基质细胞中IL-32 mRNA表达,采用Annexin V-FITC/PI双染色流式细胞术检测骨髓单个核细胞凋亡情况,采用直接免疫荧光标记全血溶血法,流式细胞术检测患者骨髓淋巴细胞及NK细胞计数情况。结果:MDS患者骨髓基质细胞IL-32 mRNA表达显著性高于对照组,RA、RAS及RCMD患者骨髓基质细胞IL-32 mRNA表达显著高于RAEB(P<0.05),而RAEB与对照组之间无显著差异。MDS组骨髓单个核细胞凋亡率显著高于对照组(P<0.05),RA、RAS及RCMD患者骨髓单个核细胞凋亡率均显著高于RAEB(P<0.05),RAEB与对照组之间无显著差异。MDS患者细胞凋亡与骨髓基质细胞IL-32 mRNA表达显著相关(r=0.870)。M DS患者骨髓中NK细胞数量与对照组无显著差异。结论:骨髓基质细胞IL-32 mRNA的表达与M DS细胞凋亡显著相关,骨髓基质细胞分泌IL-32可能是M DS骨髓细胞凋亡的原因之一,推测MDS骨髓微环境异常参与了骨髓细胞的凋亡。
Objective: To investigate the IL-32 mRNA expression of bone marrow stromal cells and its correlation w ith apoptosis of bone marrow mononuclear cells in patients w ith myelodysplastic syndrome( M DS). Methods: Bone marrow samples from 26 M DS patients and 10 iron deficiency anemia( IDA,as control) patients w ere collected,RTPCR w as used to detect the IL-32 mRNA expression of bone marrow stromal cells,and the apoptosis of bone marrow mononuclear cells w as detected by flow cytometry w ith Annexin V-FITC / PI dow ble staining. The born marrow lymphocytes and NK cells w ere detected by means of direct immunofluorescence labeling w hole blood hemolysis and flow cytometry. Results: IL-32 mRNA expression of bone marrow stromal cells in the M DS patients w as significantly higher than that of control group,the IL-32 mRNA expression of bone marrow stromal cells in patients w ith RA,RAS and RCM D w as significantly higher than that in patients w ith RAEB. There w as no obvious difference betw een RAEB and the control groups. The apoptosis of bone marrow mononuclear cells in M DS group w as significantly higher than that in the control group,the apoptosis of bone marrow mononuclear cells in patients w ith RA,RAS and RCM D w as significantly higher than that in RAEB. There w as no significant difference betw een RAEB group and control group. The IL-32 mRNA expression in bone marrow stromal cells significantly correlated w ith the apoptosis of bone marrow mononuclear cells in M DS patients. The NK cell number in born marrow of M DS patients and the control group had no significant difference. Conclusion: The expression of IL-32 mRNA in bone marrow stromal cells significantly relates w ith the apoptosis of M DS cells,and the secretion of IL-32 by bone marrow stromal cells may be one of the reasons for the apoptosis of M DS bone marrow cells. It is speculated that the abnormal M DS bone marrow microenvironment is involved in the apoptosis of bone marrow cells.
