砷化物对人胃癌细胞的抑制及放射增敏作用分析

摘要 目的探讨砷化物对人胃癌细胞的抑制及放射增敏作用。方法分别用不同浓度的三氧化二砷(As_2O_3)及不同剂量的60Coγ射线对体外传代培养的人胃癌细胞株SGC-7901进行处理。将每个细胞株分成六组,即对照组、观察A组、观察B组、照射组、联合A组、联合B组,采用荧光倒置显微镜及MTT法对细胞增殖情况进行观察和细胞计数;使用流式细胞仪对细胞凋亡情况进行检测。结果六组细胞计数、增殖抑制率、细胞凋亡率对比较,差异有统计学意义(P<0.05)。结论 As_2O_3对人胃癌细胞有一定的抑制作用,随着浓度的增加及时间的增长,其抑制作用增强,同时具有一定的放射增敏作用。 Objective To discuss the inhibiting effect of human gastric carcinoma cells and radiosensitization of people by arsenide.Methods To used the different concentration of arsenic trioxide(As_2O_3)and different dosage of coγrays for processing the human gastric carcinoma cells SGC-7901 that cultivated by subculture in vitro separately.To divided each cell line into 6groups,they was control group,treatment group A,treatment group B,exposure group,combination group A and B,to used the fluorescence inversion microscope system and MTT method for observing the cell proliferation and cell counting.To detected the apoptosis of cell by flow cytometry.Results Six groups of cells count,proliferation inhibition rate,apoptosis rate compared,there was statistical significance(P<0.05).To compared with the combination group A,the inhibition rate and apoptosis rate of the combination group B was significantly increased,there was statistical significance(P<0.05).Conclusion That has definite inhibiting effect of the human gastric carcinoma cells by As_2O_3,along with the concentration and time increased,the inhibiting effect will be enhanced,meantime that has definite radiosensitization.
