蒿甲醚对红细胞内血红蛋白释放的影响 被引量:2

摘要 为研究蒿甲醚治疗疟疾时对红细胞内血红蛋白状态的影响,用淀粉-琼脂糖混合凝胶双向电泳,分析蒿甲醚对全血及红细胞成分的影响。电泳结束后将胶板放入丽春红染色液中染色24h后,拍照留图。后将胶板烤干或室温放置待干。将干燥的胶板按联苯胺染色方法染色后拍照留图。在游离血红蛋白方面,蒿甲醚能使它向阴极移动。对疏松结合血红蛋白,它表现为HbA2现象、初释放现象,是红细胞内HbA2与HbA1结合、在与红细胞膜疏松结合,第1次通电就能脱落下来,蒿甲醚能使A2现象消失。牢固结合血红蛋白部分,它表现为再释放现象,这类血红蛋白在第1次通电时放不出来,只有再次通电时才能释放出来。蒿甲醚使红细胞成分指纹图里的10个定释带明显减弱和减少、使少量后退带完全消失。蒿甲醚能够有效干扰红细胞和全血内血红蛋白的多种存在状态,从而达到治疗疟疾的目的。 Plasmodia parasitize erythrocytes,and they need the hemoglobin inside to reproduce.The effects of artemether on the hemoglobin status of erythrocytes in the treatment of malaria were studied by hemoglobin release experiment.The effects of artemether on whole blood and erythrocyte components were analyzed by starch-agarose mixed gel bidirectional electrophoresis.After electrophoresis,the gel board was stained with the Ponceau S staining solution for 24 h,and pictures were taken.The board was baked or left to dry at room temperature,and after dry,it was stained with benzidine and then photographed for pictures.In terms of free hemoglobin,artemether can move it towards the cathode.But for loosely bound hemoglobin,it shows the phenomenon of HbA2 and initial release,and HbA2 is combined with HbA1 in erythrocytes,then combined with loose erythrocyte membrane,which can be detached after the first power on,and artemether can make the A2 phenomenon disappear.It binds firmly to the hemoglobin component,which is shown as a re-release phenomenon.This type of hemoglobin cannot be released at the first power on,and can only be released when it is electrified again.Artemether can significantly weaken and reduce10 specific release bands in the finger pattern of erythrocyte components,and a small amount of retrograde bands completely disappear.Artemether can comprehensively and deeply interfere with the various existence states of erythrocytes and hemoglobin inside of the whole blood,so as to achieve the purpose of malaria treatment.
