Practical Experience and Recent Progress of Freeze Replica Technique

根据我们的经验,简要地介绍了冷冻断裂蚀刻复型的方法和原理。详细地讨论了冰晶形成的原因和预防方法。同时也提到快速冷冻、超高真空和自动化喷镀等在仪器方面的发展。冷冻复型与放射自显影、细胞化学等结合起来将有利于促进膜结构的动态研究。

On the basis of our own experience,the procedures and the principle of the Freeze-fracture-etching-replica technique are briefly described.The cause of ice-crystal formation and its protection are discussed in detail.The developments of instrument, such as rapid freezing,ultrahigh vacuum evaporation and automation are also reviewed, combinations with radioautography,cytochemistry,etc.are expected to reinforce the application of freeze replica for dynamic study of membranous structure.
