大黄素对肾小管细胞增殖的影响及其分子机制探讨 被引量:18

摘要 观察大黄素对体外培养肾小管细胞增殖的影响。将20mg·L-1大黄素加入肾小管细胞培养液后12h,肾小管细胞增殖明显受抑,(3H-TdR掺入值2945.33±1516.24cpm·well-1Vs对照6368.17±2025.99cpm·well-1,P<0.05)。大黄素抑制肾小管细胞增殖与c-myc原癌基因和TGF-β基因mR-NA表达无明显联系。提示,大黄素抑制肾小管增殖可能通过其它调节机制起作用。 Our previous study has demonstrated mat Rheum Officinal,a chinese herb drug. can effectively retard the progression of chronic renal failure. To explore its responsible mechanism. renal tubular cells grown in cuture were made quiescent by exposure to 10g·L-1 FCS+1640 medium for 48 h, and then Emodin,an important ingredien of Rheum Offcinal,at the final concentration of 20 mg· L-1was added to culture. The proliferation of renal tubular cells was significantly inhibited 12 h after the addition of Emodin as measured by 3H-TdR uptake,while the expressions of c-myc Protooncogene and TGF-βgene mRNA remained unchange during this interval, It is suggested that Emodin might be not by means of induction of these two genes expression to inhibit cell proliferatioll.This obseration may be of help in to explain the beneficial effect of Rheum Officinal in the treatment of chronic renal failure.
