缺氧对人眼筛板细胞胶原蛋白合成影响

摘要 目的:研究缺氧对筛板细胞形态及胶原蛋白合成影响,探讨筛板细胞外基质改变与血供关系。方法:对筛板细胞进行体外培养,运用光镜及电镜观察筛板细胞在缺氧条件下形态和结构改变,采用免疫细胞化学和计算机图象分析观察筛板细胞对胶原蛋白Ⅰ、Ⅲ、Ⅳ型合成影响。结果:缺氧早期对筛板细胞形态无明显影响,但持续缺氧48小时后可发生结构变化,如胞浆内空泡增多、线粒体异形、环形样小体等变形改变。缺氧培养24、48小时,筛板细胞对胶原蛋白Ⅰ型表达量与对照组比较,差异有显著性(P<0.01);胶原Ⅲ型表达量与对照组比较,24小时显著增强(P<0.01),48小时仍有一定增高;胶原Ⅳ型表达量与对照组比较,差异有显著性(P<0.01)。结论:缺氧可增强筛板细胞胶原合成。 To study the effects of hy-poxia on the morphology of lamina cri-brosa cells (HLCCs), and the synthesis of collagen protein. Methods: HLCCs were cultured and morphological changes were studied,using both light and electron microscopes. Synthesis of collagen protein was studed, using immunocytochemistry and computer imaging analysis. Results.Hypoxia may produce degeneration of HLCCs(e. g vacuoles in cytoplasm, deformity of mitchondria and cirailar figure etc.). Change in the morphology of HLCCs war detected cells exposed to hypoxia for 24 and 48 hours demonstrated increased syn thesis of collagen I and IV type. Synthesis of collagen type III was also markedly increased after 24 hours and still remained somewhat higher than that of fhe control group after 48 hours. Conclusion. Hypoxia may increase the synthesis of collagen in HLCCs.
