摘要
目的应用免疫组化方法观察豚鼠内耳S-100蛋白的表达。方法选用健康,Preyer’s反射正常豚鼠8只,4%多聚甲醛活体灌注后耳蜗冰冻切片,采取SP免疫组化方法进行S-100蛋白染色。结果耳蜗Corti’s器内外毛细胞,Deiter及Hensen细胞呈阳性表达,基底膜下细胞未着色,螺旋缘齿间细胞,螺旋神经节细胞间纤维,螺旋韧带及血管纹均呈阳性表达。前庭毛细胞及感觉上皮下的神经纤维亦呈阳性表达。结论所及前庭末梢器官的感觉细胞及神经S-100蛋白免疫组化阳性反应提示该部位的细胞具有离子交换、分泌、吸收、保护及支持的多功能作用并与内耳神经成熟有关。
PURPOSE To observe the expression of S-100 pro-tein in the inner ear of guinea pigs using immunohistoche-mical technique.METHODS Eight healthy guinea pigs with normal Preyer’s reflex were used.The temporal bullae of the animal were serially sectioned with cryostat after in vivo per-fusion with 4% paraformaldehyde solution.The sections were stained with the monoclonal antibody of S-100 using SP immunohistochemical technique.RESULTS The S-100 protein positively expressed in outer and inner hair cells,Deiter and Hensen cells of the cochlea.The mesothelial cells of the basal membrane were negative.The S-100 protein also positively stained in the interdental cells of the spiral limbus,the fibers among the spiral ganglion cells,the spiral ligament and stria vascularis.The hair cells and nerve fibers of the ves-tibular end organ were all positive.CONCLUSIONS The fact that S-100 protein were positively stained in Cord’s organ and vestibular end organ suggested that the positive cells involved in the change of ions,secretion,resorption,protective and supportive functional roles and related to the nerve maturation of the inner ear.
