建立新生鼠脊髓细胞凋亡的动物模型

摘要 本研究目的在于制作新生鼠脊髓细胞 (神经细胞和神经胶质细胞 )凋亡模型 ,为筛选神经组织的保护物质提供新手段。为此 ,将新生大鼠一侧坐骨神经切断后 ,用无菌止血海绵包裹断离的神经近侧断端 ,海绵内加入无菌生理盐水或 NGF,术后 3、6、12 h用 TU NEL 法检测脊髓细胞凋亡状况。结果表明 ,TUNEL显示生理盐水组在三个时间段均出现大量脊髓细胞凋亡 ,而 NGF组的凋亡细胞数量很少。提示 。 An animal model on spinal cord cell apoptosis was established in newborn rats to offer a new method to sift the factors that protect the nerve tissue. The sciatic nerve of the rat born at postnatal day 1 was cut and the sponge was used to surround the proximal severed sciatic nerve, the sponge was added 10 μl NS in the control group or 10 μl(10 ng)NGF in the experimental group.At 3 h, 6 h, 12 h after operation, the L 4~L 5 spinal cord cells of the two groups were examined by terminal transferase and biotin dUTP nick end labeling (TUNEL) staining and observed in light microscope. Apoptosis of the motor neuron in NS were observed more than 25% in 3 h, more than 50% in 6 h, more than 70% in 12 h. Apoptosis of the neuroglial cell and intermediate neuron in NS were observed 40%~50% at the three timepoint. Apoptosis of the motor neuron in NGF were not observed. Apoptosis of the neuroglial cell and intermediate neuron in NGF were observed only 5%, 15%, 20% at the three timepoint. The NGF but not NS can rescue motor neuron and neuroglial cell (included intermediate neuron) at the three timepoint. An animal model on spinal cord cell apoptosis has been established in a short time after sciatic nerve is cut in newborn rat. (Figures 1~4 on plate 45)
