SIEA-Gp101分子抗原的分离纯化及其单特异性抗血清的制备

THE ISOLATION AND PURIFICATION OF SIEA-GP101 AND PREPARATION OF SPECIFIC ANTISERUM AGAINST SIEA-GP101

应用SDS PAGE、电渗、透析等方法 ,分离纯化SIEA Gp10 1分子抗原。采用多途径免疫法制备单特异性抗血清 ,琼脂扩散试验和ELISA测定抗体的效价 ,SDS PAGE和ELIB检测抗体的特异性。结果表明纯化的SIEA Gp10 1为单一抗原分子 ,对SIEA Gp10 1的抗血清进行ELIB分析 ,证明其仅识别SIEA Gp10 1抗原 ,说明SIEA Gp10 1分子具有抗原性。

Glycoprotein 101kDa molecule of SIEA was isolated and purified from SIEA with SDS PAGE,electro dialysis and additional treatments. To prepare the specific antisera against SIEA Gp101,the rabbit was immunized by the antigenic molecule. The double immunodiffusion test and ELISA were used to test the titer of the antiserum. The specificity of antibody was detected by SDS PAGE and ELIB(Enzyme linked immune blotting).The results showed that Gp101 molecule in SIEA was only recognized by the antisera against SIEA Gp101.It proved that the Gp101 extracted from SIEA only is a single molecule with antigenicity.
