摘要
为观察阿霉素所致大鼠心衰时心肌肌球蛋白重链的变化及心肌结构的改变 ,采用雄性 SD大鼠 2 2只 ,随机分为 2组 :1正常对照组 :ip等体积生理盐水 ;2阿霉素组 (ADR) :于实验开始后第 2、4天 ip ADR1mg/ kg;第 6、8天 ip ADR2 mg/ kg;第 10、12天 ip ADR3mg/ kg;第 14、16天 ip ADR4 mg/ kg,16 d累计用药剂量达 2 0 mg/ kg。停药 2 4 h后 ,称体重、心肌重量、左心室重量及心肌蛋白的提取、肌球蛋白重链 (MHC)分析 ,电镜观察心肌细胞超微结构的变化。结果表明 ADR组大鼠的体重、心体比及左室心体比 (LVM/ BW)均较正常对照组有显著改变 (P<0 .0 1)。电镜下可见心肌纤维溶解 ,肌质网扩张空泡化 ,线粒体水肿 ,嵴断裂。心肌α- MHC向β- MHC转化 ,即α- MHC含量下降 ,β- MHC含量上升。结果提示阿霉素导致的心肌损害伴有心肌细胞结构和功能蛋白质合成的改变。
To study the changes of Mysion Heavy Chain (MHC) and myocardial structure in heart failure rats induced by doxorbincin(ADR), 22 male SD rats were divided into ①control group( n =11) and ②ADR group( n =11, ip 20 mg/kg ADR given intermittently within 16 days in 8 times). Body weight of the rats, left ventricle mass to body weight ratio(LVM/BW), α MHC and β MHC were measured. The ultrastructural changes in myocardial cell were observed electromicrographically.Shifts of myosin from α MHC to β MHC occurred in ADR group in a form of the decrease of α MHC(vs control P <0.01)and the increase of β MHC(vs control P <0.01). In ADR group, an increase in LVM/BW is found (vs control P <0.01).Ultrastructural changes of myocardial cell in heart failure rats were electromicroscopically observed as lysis of myocardial filament extension and aelveoli of plasmalemma, crests rupture and swellening of mitochondria in ADR group. The results suggest that the MHC may be involved in myocardium remodeling.
