摘要
进行了海南岛3个地区的大田成龄胡椒各种外植体的组织培养试验。结果表明:只有成熟的种子才可以建立无菌培养;对在无菌环境中萌发的实生苗茎尖进行增殖培养,可获得丛生芽,再经壮苗培养和生根培养,即可得到健壮的生根植株;适合茎尖增殖、壮苗和生根培养的培养基分别为1/2MS+1.5~2.0mg/LBA+0.1~0.2mg/LIAA,1/2MS和1/2MS+0.5~1.0mg/LIAA;无菌实生苗的胚轴和叶片切段均可诱导形成愈伤组织,但不定芽的分化率极低,仅约1%。
Experiments on in vitro culture of various explants from mature p lants of black pepper(Piper nigrum Linn.)in three regions of Hainan were cond ucted.The results showed that on ly hygienic in vitro culture of mature seeds w as established.Shoot tips of seedlings germinated in vitro were multiplied.Roo ted plantlets were obtained through elongation and rooting culture of multiple s hoot buds.Media used for multiplication,elongation and rooting were1/2MS + 1.5-2.0mg /L BA+0.1-0.2mg /L IAA,1/2MS and1/2MS +0. 5-1.0mg /L IAA respectively.Callus was induced from segments of leaf and hypocotyls from seedlings,but bud differentia tion frequency was extremely low ,approximately1%.
