组培条件对不同品种芦荟试管苗玻璃化的影响 被引量:41

摘要 研究了外植体表面消毒时间、培养基中不同浓度的糖分、琼脂、BA、活性炭及培养时间等因素对芦荟组织培养中试管苗玻璃化的影响。结果表明不同品种芦荟组培苗均出现玻璃化现象且程度不同 ,以木剑芦荟最为严重 ,库拉索芦荟次之 ,华芦荟最轻 ;玻璃化苗的数量随着外植体消毒和培养时间的延长、糖分及琼脂浓度的降低、细胞分裂素 (BA)浓度的增加而加重。通过在培养基中适当调整糖分、琼脂、细胞分裂素的浓度 ,可有效地降低并控制芦荟玻璃化苗数量。添加一定浓度的活性炭 (3g/L)对芦荟试管苗玻璃化有较好的抑制作用 ,且对芦荟苗增殖、组培苗的生长、根长。 effects of different sterilized time with HgCl 2,mixing sugar concentration,agar concentration,BA concentration,active carbon concentration in the culture medium,different culture time It showed that there was vitrification in different Aloe varities The extent of vitrification was different in different varieties,Aloe arborescens was the most,then A curacao,and then A vera The numbers of vitrified plantlet increased with prolonging of sterilize time and culture time, decreaseing of sugar and agar concentration,increaseing of BA concentration The numbers of vitrified plantlets of Aloe could be controlled by the control of the concentrations of sugar,agar,and BA Active carbon with different concentration in the culture medium not only helped to control the vitrification of Aloe but improved growth,propagation,the length of root and rooting rate
