海岛棉品种根部黄萎病菌诱导表达全长cDNA文库的构建 被引量:8

摘要 以海岛棉品种 712 4黄萎病菌接种前后的根部组织为材料 ,构建的黄萎病菌诱导表达的全长c DNA文库。该文库的克隆数目为 1.15×10 7pfu,插入片段的大小在 0 .5~ 6kb之间 ,文库的阳性克隆子的比例为 84.7%。该文库可以用于抗黄萎病基因的克隆及其相关研究。在文中作者同时对棉花的文库构建所应该注意的事项进行了讨论。 A root full length cDNA library of cotton variety 7124(G.barbadense L.) was constructed after inoculation with a defoliating strain of Verticillium dahliae Kleb.. The total clone number of cDNA library was about 1.15×107pfu, its length of insertion fragment was from 0.5 kb to 6kb and the percentage of positive clones was about 84.7%. This library could meet demands of gene cloning and would be used in gene data mining. Several cautions were also discussed in this paper.
