摘要
应用免疫组织化学 SABC法对 2 4例类风湿关节炎 (RA)及 4例正常滑膜组织中血小板源性生长因子(PDGF )、转化生长因子β1 (TGF-β1 )的表达分布情况进行观察分析 ,结果发现 2 0例 RA滑膜组织衬里层的巨噬细胞样细胞、浸润的炎性细胞与血管内皮细胞、成纤维细胞可见 PDGF的表达分布 ,且巨噬细胞样细胞、浸润的炎症细胞的阳性程度强于血管内皮细胞、成纤维细胞。2 2例 RA滑膜组织衬里层与衬里下层的成纤维细胞、巨噬细胞样细胞、血管内皮细胞均见 TGF-β1 的阳性表达分布。两者比较 ,阳性细胞的染色程度、染色细胞数与分布范围大致相等。4例正常滑膜组织中 PDGF、TGF-β1 的表达分布均为阴性。认为 RA滑膜组织中 PDGF、TGF-β1 的表达分布均较正常增多且表达程度大致相等 ,两者协同作用 ,参与了 RA病理过程的滑膜衬里层增生、炎性细胞浸润。
To know the expression and distribution of platelet derived growth factor(PDGF) and transforming growth factor β 1(TGF β 1)in synovial tissues of patients with rheumatoid arthritis and to explore the probable relevance between these 2 molecules and the pathogenesis of rheumatoid arthritis, SABC immunohistochemistry assay was conducted to study the expression and distribution of PDGF and TGF β 1 in synovial tissues of 24 patients with rheumatoid arthritis and 4 cases of normal synovial tissues.The results showed that the synovial tissues of most patients with rheumatoid arthritis were observed moderately positive staining for PDGF and TGF β 1 in synovial lining layer cells and sublining layer cells including fibroblasts, macrophage like cells, infiltrated inflammatory cells and endotheliocytes,and these 2 molecules are parallel in staining extents. Whereas 4 cases of normal synovial tissues displayed no staining for these 2 molecules. These suggest that the expression and distribution of PDGF and TGF β 1 in synovial tissues of patients with rheumatoid arthritis rise distinctively comparing with normal synovial tissues and these 2 molecules have parallel staining extents., which are relevant to RA pathology including lining layer synoviocyte hyperplasia, inflammatory cells infiltration, sublining angiogenesis and synovial pannus formation.
