六亚甲基二乙酰胺对粘液表皮样癌细胞p21、PCNA、CyclinD_1表达的影响

EFFECTS OF HEXAMETHYLENE BISACETAMID ON EXPRESSION OF p21、PCNA、 CYCLIN D_1 IN HUMAN MUCOEPIDERMOID CARCINOMA CELL LINES

目的 探讨分化诱导剂六亚甲基二乙酰胺 (HMBA)诱导人粘液表皮样癌细胞后 ,p2 1、增殖细胞核抗原 (PCNA)和CyclinD1 表达的变化。方法 用 2mmol/LHMBA对培养的粘液表皮样癌细胞进行体外诱导 72h后 ,分别采用免疫组织化学、图像分析等方法对处理与未处理细胞的p2 1、PCNA和CyclinD1 表达变化进行定量分析。结果 HMBA处理的粘液表皮样癌细胞p2 1表达明显高于对照组 (P <0 .0 1 ) ;PCNA和CyclinD1 表达明显低于对照组(P <0 .0 1 )。结论 HMBA通过影响p2 1、PCNA和CyclinD1

Objective To study the alteration of p21、PCNA and CyclinD 1 protein after mucoepidermoid carcinoma (MEC-1) cells were induced by Hexamethylene Bisacetamide (HMBA). Methods MEC-1 cells were induced 72 hours by 2mmol/L HMBA in vitro. Proteins of p21、 PCNA and CyclinD 1 were detected using immunohistochemistry. The positive degree of p21、 PCNA and CyclinD 1 was analyzed using image analysis. Results The expression of p21 in HMBA groop was higher than control. The expression of PCNA and CyclinD 1 in HMBA groop was lower than control. There were significant difference between the treated and the non treated group ( P <0.01). Conclusion HMBA can inhibit proliferation of MEC-1 cells by increasing the protein of p21 and decreasing the proteins of PCNA and CyclinD 1.
