摘要
目的 :研究干扰素 α(IFN α)对肝纤维化大鼠肝星状细胞 (HSC)凋亡的影响。方法 :用四氯化碳(CCl4)复制肝纤维化模型大鼠 32只 ,随机分为A、B、C 3组。A组 :6只 ,造模结束即处死 ;B组 14只 ,皮下注射IFN α 10万U 10 0g 8周 ,C组 12只 ,注射等量的生理盐水 8周。治疗结束时用TUNEL法检测各组肝星状细胞凋亡情况 ;HE染色观察肝组织学变化 ;免疫组化染色检测中间丝蛋白 (Desmin)、α 平滑肌肌动蛋白 (α SMA)、I型胶原、转化生长因子 1(TGF β1)等指标。结果 :B组较其他两组肝组织学明显改善 ,肝纤维化程度减轻 ,I型胶原、TGF β1的显色指数降低 ;B组肝星状细胞明显减少 ,凋亡指数增加 ,与C组相比 ,差异有统计学意义。结论 :IFN α在体内可诱导HSC凋亡 ,这可能是IFN α抗肝纤维化的作用机制之一。
Objective:To study the effects of interferon α(IFN α) on the apoptosis of hepatic stellate cells(HSC) of rats in vivo.Methods:Thirty two rats in which liver fibrosis model was induced by carbon tetrachloride(CCl 4) were divided into three groups. Group A: 6 rats were put to death after CCl 4 had been given for 8 weeks; Group B:14 rats received 0.1MU/100?g of IFN α for 8 weeks by subcutaneous injection; Group C: 12 rats were given an equivalent dose of saline. Cell apoptosis was calculated by TUNEL and histological examination was also performed . The expression of Desmin,α SMA,collagen type I and transforming growth factor β 1(TGF β 1) was measured by immunohistochemical assay.Results:The degree of hepatic fibrosis was markebly lower in croup B than in croup C,and coloration indexes of collagen type I and TGF β 1 were significantly lower than those in other two groups.The number of HSC in croup B reduced markedly and the index of apoptosis increased with significant difference when compared with croup C .Conclusion:IFN α can induce the apoptosis of HSC in vivo and this may be one of the michanisms of IFN α to counteract liver fibrosis.
