整合素a_5亚单位在大鼠角膜碱烧伤愈合中的表达 被引量:1

摘要 目的 观察碱烧伤后角膜创面上皮愈合过程中整合素 α5亚单位的表达 ,探讨整合素 α5在角膜上皮损伤愈合中的作用。方法  Wistar大鼠 1 8只 ,随机分为 6组 ,每组 3只 ,其中 1眼为碱烧伤模型 ,另 1眼为自身对照眼。对制造模型眼后 0、7h、1、3、7、1 4d的角膜标本 ,HE染色行组织病理学检查、间接免疫荧光和免疫组织化学染色观察整合素 α5亚单位的表达与分布。结果 伤后 7h,邻近上皮细胞开始表达 α5亚单位 ,伤后 1、3、7d,角膜上皮层显示 α5表达逐渐增加 ,差别有显著性 (P<0 .0 5)。伤后 1 4d,实验组表达仍呈上升趋势。结论 整合素 <Abstrcat>ObjectiveTo observe the expression of integrin α5 subunit during corneal alkaline burn wound healing in rats. MethodsEighteen Wistar white rats(body weight 200~250g) were divided into six groups. Three rats in each group were made into corneal alkaline burn model by 0.5mol·L-1 sodium hydroxide solution. The cornea were surgically removed at six different time points(0?7h,1?3?7?14d)after operation, three rats for each time. The localization and expression of integrin α5 subunit was determined by indirect immunofluorescence and immunochemical methods. ResultsThe expression of integrin α5 subunit of experimental groups increased in alkaline burn cornea at seventh hour after operation, and it increased gradually after 1day,3,7,14days.ConclusionThe expression of integrin α5 increased markedly gradually during corneal alkaline burn wound healing, as promoted the adhesion of cellcell and cellECM.
