γ-干扰素基因修饰人肝癌细胞的体外生物学行为

摘要 目的 研究γ干扰素(IFNγ)基因修饰人肝癌细胞的某些体外生物学行为。 方法 用逆转录病毒载体建立4种γ干扰素基因修饰人肝癌细胞,分别检测外源基因表达、细胞形态结构、体外生长速率及60Co照射的影响。 结果 4种IFNγ基因修饰人肝癌细胞表达不同生物活性水平的IFNγ,体外生长速率有不同程度的下降。经60Co照射及冻存处理,复苏后仍能分泌较高水平的IFNγ。 结论 IFNγ基因修饰人肝癌细胞体外生物学行为有一定程度的变化。 PURPOSE To study the in vitro biological activities of hepatocellular carcinoma(HCC) cell lines following transduction with γ interferon gene.METHODS Four IFN γ gene transduced HCC cells were established using retroviral vector. IFN γ gene expression,morphology of transduced cells, in vitro growth rate and the effect on gene expression after irradiation and cryopreserevation were analyzed.RESULTS Different IFN γ transduced HCC cells secreted different amounts of IFN γ, and showed decrease in their growth rate. The capacity of producing IFN γ in transduced cells was retained after irradiation and cryopreservation.CONCLUSION Some changes occured in transduced HCC cells,which can act as potential tumor vaccine.
