摘要
目的 研究ER阳性和ER阴性的人乳腺癌细胞株整合素亚型α5β1、α4β1和组织蛋白酶D蛋白的表达和细胞生物学特性的关系。 方法 采用细胞培养、Southern杂交、Northern杂交和免疫组织化学技术,检测ER阳性MCF7及ER转染阳性MDAMB231和ER阴性MDAMB231人乳腺细胞中整合素亚型α5β1、α4β1和组织蛋白酶D蛋白的表达,了解它与细胞生物学特性的关系。 结果 MCF7、ER转染阳性MDAMB231和MDAMB231细胞中整合素亚型α5β1基因DNA,mRNA表达量各不相同。整合素亚型α5β1、α4β1和组织蛋白酶D蛋白在三株人乳腺癌细胞中的定位各不相同,且存在数量上的差别(P<005)。三苯氧胺具有诱导α5β1表达的作用,而对α4β1和组织蛋白酶D无影响。 结论 雌激素受体的表达和整合亚型α5β1、α4β1和组织蛋白酶D状态有关。
PURPOSE To study the experssion integrin α 5β 1、α 4β 1 and cathepsin D in ER positive and ER negative human breast cancer cell dlines and their biological signifence. METHODS The DNA and mRNA status of integrin α 5β 1 was examined by Southern and Northern blotting in MCF 7,MDA MB 231 and ER transfected MDA MB 231 human breast cell line,The expression of integrin α 5β 1,α 4β 1 and cathepsin D proteins were tested in these three cell lines with or without tamoxifen interference by immunochemical staining.RESULTS Integrin α 5β 1 mNRA status is different in three cell lines; Integrin α 5 protein locallizes in the cellular memberance,whileas α 4β 1 and cathepsin D in cytoplasm,The expression of integrin α 5β 1 in ER positive MCF 7 and ER transfected MDA MB 231 cell line is higher than that in MPA MB 231. Tamoxifen induced integrin α 5β 1 experssion ( P <0 05),no signifience on α 4β 1 and cathepsin D gene expression. CONCLUSIONS The ER staus of breast cancer cell lines is related with the expression of integrin α 5β 1,α 4β 1 and cathepsin D,and tamoxifen inhibits the metastatic capability of breast cancer cell lines.
