贡柑珠心胚培养与微型嫁接研究 被引量:1

摘要 选取四会贡柑珠心胚,作离体培养。比较了几种激素的配比和水解酪蛋白(CH)对珠心胚发育、珠心苗子叶分化的效应,以及培养基的状态对胚发育、珠心苗茎切段的影响。实验表明:MT+IAAlmg/l+BA0.2mg/l+CH400mg/l和MT+BA0.5mg/l+ZT0.5mg/l两种培养基对珠心苗诱导效果较好。前者在IAA参与下,珠心苗下胚轴切段芽的分化随BA浓度增加而提高;后者BA和ZT同时存在,对子叶分化产生小芽有明显的促进作用。液体培养能加速小苗的生长速度,增加小苗数量。取两周苗龄的江西红桔实生苗为砧木,珠心苗为接穗,无菌条件下,于试管内进行微型嫁接,嫁接苗培养在1/2MS培养基中,成活率达95%以上,两周后转移沙床,注意保持湿度,成活率达90%以上。 Nucellar embryos from Si-Hui "gonggan" (C. reticulata cv. yu dong) matured seeds were cultured in vitro. Effects of several hormones and CH on the development of nucellar embryos and the cotyledor differentiation were studied.Two mediums, MT (Murashige-Tucker) containing lmg/l IAA, 0.2mg/l BA and 400mg/l CH, and MT supplemented with 0.5mg/l BA and 0.5mg/l ZT were found to be effective to induce differentiation. On the medium containing IAA and varying BA concentrations, as BA concentrations increased, the abillity of cultured sections of hypocotyl of the seedlings to form buds increased; the medium containing both BA and ZT was more effective for the bud formation of cotyledons. Liquid culture could speed up growth and a large number of seedlings were produced. The research of micro-grafting used Giang-Xi "hongi" (C. reticuiata Blance) (two weeds old) as the stock and the nucellar seedlings as the scion. On 1/2MS (Murashiga Skoog)mudium, the percentage of survival seedling-grafting was over 95%, and after transplanting, the survival rate of seedlings was over 90%.
