摘要 将转基因植物组织提液与底物[r-32p]ATP和卡那霉素进行反应,再将磷酸化的卡那霉素转移到P81磷酸纤维素离子交换纸上,放射性标记的卡那霉素可通过斑点放射自显影进行观察。此法简便、快速,灵敏度较高。 This paper described a method of dot autoradiography for the detection of neomycin phosphotransferase in crude cell extracts. The crude cell extracts of transgenic plants reacted upon the substrates 〔r-32P〕 ATP and kanamycin at 37℃ in tubes. After the enzymatic reaction, the phosphorylated kanamycin was transferred to P81 phosphocellulose ion exchange paper, and the radiolabeled kanamycin was visualised by dot autoradiography. This method is simple, rapid and sensitive.
