用cDNA芯片技术快速鉴定鼻咽癌组织消减文库中的差异基因 被引量:1

摘要 迅速分离、鉴定鼻咽癌组织中差异表达基因是研究其发生、发展分子机制的基础.先用正常鼻咽组织和鼻咽癌组织进行抑制性消减杂交,构建了消减文库再用含有 14 000个基因片段的cDNA芯片,对此库进行了筛选,结果发现了417个差异表达的基因,其中在鼻咽癌组织中上调的170个,下调的247个.抑制性消减杂交提供了一个差异基因的富集库,而利用cDNA芯片同时筛选上万个基因; Rapid Identification of differentially expressed genes is basic work for understanding molecular mechanism of pathogenesis of NPC. First, subtractive libraries were constructed using suppression subtlactive hybridization (SSH) of NPC tissue and noed nasopharyngeal tissue. Then, these two li- braroes were screened by cDNA chip which contains 14000 genes. The results show that total differentially expressed genes are 417, among which in NPC tissue upregulated genes are 170 and downrgulative genes are 247. It is suggested that many differentially expressed genes can be simultaneously gained by combination of SSH and DNA chip.
