摘要
将20只短肠SD大鼠随机分成单纯TPN组及GIn-FPN组,行肠粘膜上皮细胞增殖。凋亡及相关基因表达的检测,探讨谷氨酰胺(glutamine,Gln)促进TPN短肠大鼠小肠代偿的分子机制.结果表明:单纯TPN可造成大鼠小肠粘膜明显萎缩,添加Gln的TPN可显著缓解肠粘膜萎缩.其肠粘膜上皮细胞PCNA表达显著增高,凋亡指数下降.Gln抑制细胞凋亡的作用与bcl-2表达上调及bax表达下调密切相关.
To understand the underlying mechanism of the adaptation of the remnant small intestine in rats receiving glutamine, 20 parenterally fed, short bowel SD rats were randomized divided into simple TPN group and GIn-TPN group, the expression of proliferating cell nuclear antigen (PCNA), the occurrence of the apoptosis and related gene expression were determined. Our results show that Atrophy of the remnant small intestine was occurred TPN group, however it was significantly alleviated in GIn-TPN group with increasing of PCNA expression and decreasing of the rate of apoptosis, UP-regula- tion of apoptosis -related bcl-2 gene and down-regulation of bax gene plays an important part in de creasing of cell apoptosis.
